I have heard about global warming before, probably back in g rade school but I knew a little about it or I only knew the meaning of the term global warming. The documentary gave me a deeper understanding of what global wa rming really is and what will be the consequences if we do not do something abou t it. The greenhouse gases are caused by producing too much CO2. When Al Go re compared the old and new photos of the same place, the difference was very ob vious. I can clearly see that the glaciers are starting to melt. Al Gore also sh owed graphs and charts showing that every year the production of CO2 is getting higher also earths temperature is getting warmer. Other nations are trying to do something about global warming. Most national gov ernments have signed and ratified the Kyoto Protocol. The Kyoto Protocol is an a mendment to the international treaty on climate change, assigning mandatory emis sion limitations for the reduction of greenhouse gas emissions to the signatory nations. However, as in English, there are other ways of telling about future events. In the making of Avatar, unlike in the making of Jurassic Park, the technology was developed specifically for the development of the movie. Chemicals Needed for Muscle Contraction The goal for this lab was to determine the conditions that demonstrate what chemicals in muscle fibers are necessary for contraction and which ones prevent muscle contraction from occurring in a simplified system in order to determine the minimum requirement for contraction. Methods In order to prepare muscle fibers, a single thread was obtained from a mass of glycerinated muscle fibers which was about 0.5 mm in diameter. The muscle was a rabbit psoas muscle in 50% glycerol (stored at -10 degrees C) obtained from Carolina Biological. In order to have an observation of what was taking place with the muscle, Nikon E400 was used. The fiber was placed on a slide in the presence of 0.05 M KCl and 0.005 M K phosphate buffer (pH 7). The first step was to detect the minimum requirement solution for muscle contraction to take place. This was done by using the following solutions: 0.001 M MgCl2, 0.001 M CaCl2, and 0.1 M ATP. The requirement was determined after observing the changes taking place when each of the solutions were added to the muscle separately, in combinations of two, and all three solutions together. After the minimum requirement was determined, chelators were used to see whether they inhibited contraction in the presence of the solution that caused contraction. The chelators were 0.002 M EDTA and 0.002M EGTA. The EDTA binds Ca ++ and Mg ++. EGTA binds only Ca++. The chelating agents enhance the solubility of magnesium and calcium and allow them to spread out of the muscle fibers. This causes the removal of the ions from the actin and myosin environment. It was important that the chelators were added before the contracting agents when inhibition was being tested. Otherwise, it would be impossible to detect any changes once the contraction has taken place; contraction is not reversible once it occurs in a simplified system. Then Solution A and Solution B were used to examine the localization of myosin and actin in the myofibrils. Solution Aà ¢Ã¢â€š ¬Ã¢â€ž ¢s function was to solubilize and remove myosin in the form of monomers from its loci which is in the myofibril. Solution B had the same behavior as the actin. The Solutions were placed on the myofibril to see changes that took place and observations were made. Discussion and Conclusion During this lab, using the microscope we examined the changes that took place when certain solution were introduced to the rabbit psoas muscle fibers. The solutions caused contraction, inhibited contraction to occur, or had no effect on the sarcomeres at all. We used glycerinated myofibrils from rabbit psoas muscle which is a type of striated muscle. Rabbit psoas muscle was a good model to use for this lab since the fibers are long and straight. Also one other advantage was that there were not a lot of connective tissues connecting the muscle fibers together. This was an in vitro model meaning that experiment was completed outside the living organism. The Phase Contrast with magnification of 10X/ 40X was used during this lab to examine the slide because the cells are transparent and Phase Contrast is the best option to use in order to have a good resolution. Under a microscope the myofibers were striated and they had a repeating pattern of bands and lines. The pattern was caused by parallel organization of protein filaments within the myofibrils. In the myofibril, there are two types of filament- the thick filaments which consist of the protein myosin and thin filaments composed of the protein actin. As demonstrated on Table 1, Mg2Cl2, Ca2Cl2, and ATP were the solution used to determine the changes taking place with the muscle. All of the three solutions were placed on the slide which consisted of a thread of muscle fiber and contraction of the muscle was observed. We could tell when contraction was taking place because the fiber was short in length and it was easy to see the changes such as the shortening in length and the color change. When Mg2Cl2 and Ca2Cl2 were added individually to the fiber, nothing happened. However, as soon as the ATP was placed on changes were easily observed. ATP caused muscle contraction by itself. The sarcomeres in the muscle fiber shortened in length and the color changed from light yellow to darker yellow. However, in order to make sure this was the minimum requirement for muscle contraction, we added Mg2Cl2 with ATP and Ca2Cl2 with ATP. With the MgCl2 and ATP, the contraction occurred immediately as the solutions were added. The contraction was even faster than the ATP alone. Then ATP and Ca2Cl2 solutions were introduced and this also caused contraction. Even though the combination of the two solutions caused contraction to occur faster than ATP by itself, it was slower than the solutions of ATP and Mg2Cl2. As a result, we concluded that ATP was the minimum requirement needed for the cells to contract. All the solutions that caused contraction were not in one dimension because every component of a sarcomere was facing changes except the A band which stayed the same. The I bands, the M line, the Z lines, and the actin and mysosin- they were all decreasing in length in order to cause contraction. At the end, it was determined that ATP was the requirement for glycerinated muscle contraction. When there is no ATP present, the myosin heads in the muscle will not be activated and it would not bind to the actin. In glyceri nated tissues, the combination of KCl and MgCl2 with ATP increased the strength of muscle contraction. This was mainly due to myosinà ¢Ã¢â€š ¬Ã¢â€ž ¢s high affinity for these ions. Table 2 shows whether contraction was inhibited in the presence of chelating agents, EDTA and EGTA, when it was used with the main solution that caused contraction, ATP. EDTA and EGTA did not inhibit contraction from taking place, but the contraction was slower than when ATP was present. EDTA is a chelating agent that binds Ca++ and Mg++ and EGTA is a chelating agent that binds Ca++. The chelating agents increase the solubility of Mg 2+ and Ca2+ so that they can leave the muscle fibers. With ATP and the chelating agents, contraction occurred and there was no inhibition taking place. Table 3 shows two different solutions, Solution A and solution B, and their effect on actin and myosin. As shown in the table, solution A had KCl, phosphate buffer, Na pyrophosphate, and MgCl2 while solution B had phosphate buffer. Both of these solutions did not cause any contraction in the muscle based on our observations. However, changes were observable because in both cases the fibers changed color; they became lighter yellow. This meant that the muscles were not contracting. Solution A made the mysosin more soluble and solution B acted in the same manner as the actin. When comparing the muscle in living tissue, the glycerinated muscle system is different. The glycerination technique eliminates ions and ATP from the tissue and disrupts the troponin/tropomyosin complex. When the complex is interrupted, the available binding sites on the actin fibers are no longer blocked ( Cell and Molecular Biology). As a result, Ca2+ is not one of the requirements to cause contraction. On the other hand, since there is no ATP is in the glycerinated tissue, the myosin heads cannot be activated to cause contraction. After the muscle contracted it did not relax since there were no opposing muscles to pull it. Also, muscle fibers do not contract when there are no stimulations or nerve signals and this was one of the differences with glycerinated muscle and the living cell muscle. Errors could have occurred in this lab if one used very thick muscle strands. Having thinner strands were better to have good results. Also, it is possible that much of the calcium was still in the sarcoplasmic reticulum of the glycerinated muscle, which would have lead into incorrect results. All in all however, the lab was successful and we have obtained what we were looking for- what solutions cause contraction, which ones inhibit, and what is the minimum requirement for muscle contraction.

The main purpose abortions are immoral is how they are so viciously done. Everyday, innocent, harmless fetuses that could soon be laughing children are being brutally destroyed. One form of abortion is to cut the fetus into pieces with serrated forceps before being removed, piece by piece from the uterus by suction with a vacuum aspirator. Another form consists of bringing the fetus feet first into the birth canal, puncturing its skull with a sharp instrument and sucking out the brain tissue. A Narrow Escape Essay

This is a true story. It is the story of our narrow escape from sudden death. My brother who is handicapped & myself went to Dadar in Mumbai to visit an Aunt. She lived very near the beach. I was just 15 then & my brother 5. We loved going to the beach but normally always went with our mother or elder sister. On our way back, temptation got the better of us & we came up with the great idea of walking along the beach instead of taking the taxi back home. If we walked along the beach we could get onto the road at the Mahim Fort some distance away from Dadar, from where we could take a taxi to our home. All this seemed fun. We were very excited. My brother totally agreed with my idea. However, we did not tell our aunt about all this, as she might refuse. She thought we were going to take a taxi back home. So off we ran to the beach, once our aunt closed the door. On the beach, we got playing on the sand, making little sand castles, collecting sea shells of all shapes & sizes & time & again dipped our legs in the water enjoying the waves that lashed against the shore. We wrote our names on the sand only to be erased by the coming waves. The tide was coming in but we were blissfully unaware of anything. We enjoyed ourselves thoroughly. We had a great time. We drank coconut water, ate bhel puri & leisurely walked along in the sand. My brother has a paralyzed right hand & walks with a limp. But nothing came in the way of his enjoyment on the beach that day. We splashed water on each other’s faces. I ran and he tried to catch me. We had so much fun. Time passes so quickly when one is happy. Our games continued. We never knew when afternoon turned to evening. It was beginning to get dark now and it was then that we decided to move on. Engrossed in play we had forgotten to look around. We moved ahead slowly, not noticing the tide. We walked on crossing the rocks and boulders. As we moved ahead, at one point, we were shocked to see that we could not go any further as the waters had covered the place ahead. The tide had come suddenly. Gripped with fear we turned back. But to our astonishment the waters behind us had also touched the shore. Now we were left on a small dry patch of land & were surrounded by water on all sides. We could not move forward, nor go back. There was nowhere to go. Now we realized that we had landed ourselves in a great danger. We could see the Mahim Fort some distance away; the waves lashing against it. That was the place from where we had planned to exit. We stood there confused, afraid and shocked. Alas, we found there was no way out. It seemed our mind has stopped working. We were terrified. In desperation we looked around for some way out. Our only hope was a tall wall that was before us & there were huge boulders near the wall. But how could we climb the wall ? It was dark & no one in sight. The water level was increasing and threatening to cover this place too. Everything was happening so fast. We had very little time left with us. My younger brother was frightened and started crying. I tried to console him, but I myself was also much terrified. What next ? It seemed like an impossible situation. Escape seemed more & more difficult. Only God could save us. So we began praying desperately for God to work a miracle & send His angels to save us. I started cursing myself for not being careful. Due to my folly, we had landed in such a terrible situation. We called for help with the hope that someone would be around that high wall. The water came & brushed our feet pulling us backward into the sea. Would we have a watery grave ? The sea which looked so pleasant and beautiful in the day now looked wild and dangerous, eager to devour us. I thought, what will happen to my parents and sister? They will be devastated. Probably, nobody would know about us or find us. But I restrained my thoughts and decided that I must do something. With great effort, I managed to get my brother onto a huge boulder. He was safe, at least for the time being. The water was now rising. It had reached a little above my knees & I clung onto a boulder for dear life. We were shivering with fear, but we did not give up. We shouted for help, but it appeared there was no one to help us. I decided to get my brother across the wall by myself. I picked him up and made a desperate attempt to get him across the wall, but in the process we slipped and he barely managed to prevent the fall from the boulder into the surging water. His legs got a bit hurt and he cried in agony and pain and fear. I realized that on our own, we can never cross that big wall. Now only help from outside could save us, and there seemed to be none. The ever rising water level was making us frightened more and more. In our desperation, we shouted in a chorus for help. Then miracle happened. Suddenly, we saw two men looking from over the wall. They seemed very scary, rugged, rough & tough guys. But I knew that this was help sent from God in answer to our desperate prayers. They were also shocked to see us in that condition. I pleaded with them for help & they were astonished to see us out there almost drowning. At first, they stood there hesitated. They knew that it was dangerous and they had to risk their own lives to help us. But then they decided to come to our aid. It was not an easy task. Time was running out for us. We were drowning but God was with us. The men came down the wall slowly and carefully & along the boulders. One mis-step and they would fall into the water. But, they moved very carefully and finally succeeded in getting hold of my brother & carried him to safety over the wall. I felt a sense of relief & gratitude. I felt tremendous joy to see that my brother was safe and across that wall. For a moment, I forgot that my own life was still in danger. Now I made every conscious effort to climb the boulders. The men were strong and they pulled me up. With their help, I got over the wall. I got some bruises in my hands and legs, but who cares, since we were alive and safe. Once up, they screamed at us for being so careless & warned us never to do this again. We had learnt our lesson. Overwhelmed with gratitude we thanked the men for their timely help & also tried to give them some money. But they refused to accept that. They too were happy that they were able to save us. They had taken a risk to come down the high wall & walk over those boulders which could have given way & caused some more trouble. Those two men, appeared to be rough guys, but for us, they were like angels, who came as if from nowhere, and saved us from a certain death. They were kind and risked their own lives to save us. The men then stopped a taxi for us. We thanked them once again and hoped to meet them sometimes. We got into the taxi and reached home. Scared & terrified by what we had just experienced we looked like old turkey buzzards. We were wet and bruised, and heaved sigh of relief that we had reached home safely. We narrated all the events to our parents. They were speechless and could not believe what we said. At first they were angry & upset and scolded us for our carelessness, but also happy to see us safe. My mother cried and hugged us and made us promise never to repeat this mistake ever again. We praised & thanked God together for saving our lives. It felt so nice to be back to safety in our home with our parents. All this seemed like a miracle. We could not believe that we had been through a near catastrophe. It was indeed a very narrow escape. This is a lesson for all children who venture on the beach to be alert & watch the tide coming in & come back to safety in good time. Luck may not always be on our side. Rather one should be careful and avoid taking unnecessary risks. Life is precious and one should not waste it by sheer negligence. We got our lesson that day. I just thought that my life will be ended anytime by now. A few moments later, I didn’t feel anything. Slowly I opened my eyes and saw a blue van was just right in front of me. The speech that Randy Pausch delivered was one-of-a-kind since it made the world stop and pay attention and this was possible because in the cause of the lecture, Pausch was upbeat and humorous. He alternated between wisecracks, insights on computer science and engineering education (Randy). He offered inspirational life lessons followed up by actions such as performing push-ups on stage. This makes the listener wonder and be encouraged of the fact that despite Pausch knowing that he is going to die and he is in his final hours of life, he is still jovial. It was a way to explicitly encourage listeners on how to carry on with life despite knowing that we will die later on. On the other hand, his book advises individuals on building multi-disciplinary collaborations, working in groups and good interaction with other people. Pausch expresses a five star attitude because despite being diagnosed with terminal cancer, he explains that it is just an engineering problem. He notes that he had only 3-6 months to live and with it he has three small children who would never know him. The book draws peoples emotion without the inclusion of paralinguistic features and can at times subject them to subjective opinions. The lecture he gave helps people know how to live your life as if you were dying, an irony he put out well. He avoids showing deeper emotions in such a way that it made the lecture a celebration of his life. We are made to know what to do when life throws brick walls on our way. He said that we have a choice which is, to stop or go around the bricks. We are supposed to play out our cards to get dealt instead of bitching about them. This is explained in details in the book as compared to the lecture he gave. The deeper explanation encourages one to go around the bricks than stopping. On the other hand the lecture in explaining obstacles, expresses death with an example about President Cohen, who having realised Pausch was going to do this talk, said to him that he should introduce a concept about having fun (Randy). He explains that just like a fish talking about the importance of water he also did know how to have fun despite waiting for his death. He explains that he is going to con tinue to have every day he is left with because theres no other way to play it. In the book, Mr. Pausch says to us that "The brick walls are there for a reason, they are not there to keep us out, but the brink walls are there to give us a chance to know how badly we want something (Pausch and Jeffrey, 51). We always find out that a lot of people want some shortcut in life but through reading the book, there is a candid explanation that shows that the best shortcut is the long way, this boils down to two words as hard work. It explains that if only we could stop taking a shortcut and work a little harder then we can have a better life. The reference to the brick wall has been made several times to remind us that it is so true. The only way to live with the obstacles is that as he says, "Time is all you have. And you may find one day that you have less than you think (Pausch and Jeffrey, 111). This is a major realization in anyones life since many people think that they have so much time, but it slides by so quickly. We should take the time to do the things we wan t and never put them off since "Luck is indeed where preparation meets opportunity" (Pausch and Jeffrey, 119) we should always be prepared. The lecture has a positive voice, making it interesting and helpful. Knowing that you are going to die can be at times devastating because any news about an ill heath that would shorten life can be quite disturbing. To add on it one is left hopeless to find that even the doctors cannot do anything to help. On the contrary, Pausch experiences and the last lecture he gave were made up of true and touching incidences which help live positive or find something that will help us want to be a better person. There are some bits of wisdom that can be carried and copied into our lives. Despite involving oneself in the emotional moments, we are also forced to laugh several times too. The lecture touches anyone who has had their life touched by cancer, it benefit all from the optimism Mr. Pausch has because of the seriousness of the issues at hand. The optimism in the current negative world is very uplifting. In contrast it is from the book that we are able to understand that experience is what we get when we dont get what you want in life. We are made aware of this when Mr. Pausch was describing the strength and love he had for his wife, Jai. He says that she is the one who will be left to raise the three children to preschool-age (Pausch and Jeffrey, 130). As a matter of encouragement to the readers, he talked about how his cancer diagnosis had encouraged him and given him the time to have open and vital conversations with his wife. This he says would not have been possible if he still had a lot of time to live. Through all the encouragement he gave her, I believe made he cope with the fact and as he says, she was coping and was doing it without the attention and even adulation that surrounded the last days of his life. This cannot be so in the daily life of an individual surrounded by such obstacles. In this situation, weak individuals would have circum to sever stress or any other dis ease that would come as an emotional outburst. The reality and power expressed in the book, gives us a lot to think about in what we want to prioritize in our lives. It directs what we might say to the people we love when we know that we are going to leave them behind. It arouses the desire to want to kiss your friends or your parents and call relatives too because he sums that "All you have is what you bring with you" (Pausch and Jeffrey, 159). Finally, to enable the message reach the audience, we are able to see that throughout the lecture, professor Pausch remained very upbeat, he was charismatic and the nerdy persona he displayed was infectious. Fro the book, as we read, we cant help but we consider and ponder what we would when faced with such circumstances if we know we have only months to live. With good communication and savvy and personality, he avoids very reserved, sombre tone for this speech since it would treat every word as if it were a matter of life or death. This method would have drawn more attention to his condition rather than the core message and that is why everyone who listens to the message is left with the desire to change. Even though he was being personal to some extend and the entire speech having a content with personal lessons learned through life illuminated through personal stories, Randy had a way to tell the stories while still creating some distance and remaining somewhat clinical (Randy).

The significance of the story’s tragic end is that the desperation and fury of the oppressed are not of any consequence, as is indicated by Clara’s unwept death or the kind of end that Hoink, Ganz and Butch eventually meet with. The personal good and bad traits of the characters notwithstanding, the y have a common source of motivation – lack of options – that drives them towards planning and executing (often unsuccessfully) criminal activities. Modern theories on self-improvement suggest that if one thinks one can do, one can. Examined from the perspective of Butch’s experience, the validity of such theories becomes debatable.
